Abstract

Background and Purpose Diabetes Mellitus (DM) is a metabolic disease that developed due to the pancreas does not sufficient to produce insulin or the body cannot effectively use the insulin it produces. Genetic factors have an essential role in the development of Type 2 Diabetes Mellitus (DMT2), which impaired insulin production by pancreatic β cells, insulin resistance, and action. The single nucleotide polymorphisms in the KCNJ11 affected the pancreatic β cell activity that can inhibit insulin release, thus causing a decrease in therapeutic effectiveness. Clinical manifestations of type 2 diabetes are influenced by environmental and genetic factors, and lack of physical activity and increased nutritional intake are the main causes of type 2 diabetes. Method This type of research is descriptive. The research was conducted at the Molecular Biology Laboratory, Surakarta National High School of Health Sciences in February-March 2023. The research subjects included 25 prolanis members of Type 2 Diabetes Mellitus at the Bulu Sukoharjo Health Center, who were taken using a total sampling technique. Data were collected by filling out informed consent, the detection of the KCNJ11 gene was examined by the PCR method. Conclusion Based on the results of this study it can be concluded that the detection of the KCNJ11 gene in 25 blood samples of patients with type 2 diabetes mellitus had been carried out by PCR with the amount of DNA template 5µl, annealing temperature of 58°C using reverse primer 5'- ACG TTG CAG TTG CTT TTC TT 3' , forward primer 5'- GAC TCT GCA GTG AGG CCC TA-3' at a target of 210 bp resulted in 100% amplification of the KCNJ11 gene.
