Abstract

Asian swamp eel is a freshwater fish that contains collagen, especially in the skin and bones, but the application is still not optimal. Generally, collagen is mostly obtained from cows and pigs, where there are concerns about infectious and religious issues. The study aim to isolate collagen and determine the characteristics of collagen produced from the skin and Asian swamp eel bones. Collagen isolation was carried out in 2 stages, namely the pre-treatment stage and chemical hydrolysis. Pre-treatment optimization was carried out with 3 variations of treatment by immersion in 3 different concentrations of NaOH, namely a concentration of 0.05 M; 0.1 M; and 1 M. The pre-treatment was carried out for 6 hours with replacement every 3 hours for skin and for 24 hours with solvent replacement every 6 hours for bone. Chemical hydrolysis was carried out by soaking the pre-treatment results in 0.5 M CH3COOH solution for 6 hours, then the salting process was carried out using 0.9 M NaCl. The yield was carried out by chemical and physical characterization. The optimization results from 3 variations of NaOH concentrations obtained collagen yield of 0.07% at a concentration of 0.05 M NaOH; 1.18% at 0.1 M NaOH concentration; 0.14% at a concentration of 1 M NaOH. The most yields were obtained at a concentration of 0.1 M with a fluffy white color. The characteristics of the yield were proximate components such as water content of 8.35%, fat content of 12.93%, and protein content of 21.5%. FTIR results of collagen from skin and bone Asian swamp eel showed typical collagen absorption peaks, namely Amide A (3299.25 cm-1), Amide B (2937.5 cm-1), Amide I (1653.90 cm-1), Amide II (1538). , 16 cm-1), and Amide III (1242.08 cm-1).
