Abstract

Pseudomonas aeruginosa is a bacteria that causes diabetic ulcers in diabetics. This study aims to determine the antibacterial activity of ethanol extract and fraction of banana weevil against Pseudomonas aeruginosa bacteria. Banana weevil extraction in this study used maceration with 96% ethanol then dissolved in warm water and continued with fractionation using n-hexane and ethyl acetate solvents. The results of extraction and fractionation were tested for antibacterial activity against the extract, n-hexane fraction, ethyl acetate fraction and water fraction of Kepok banana humps using the diffusion method with concentrations of 25%, 50%, and 100%. Clinical comparators used the Clinical and Laboratory Standards Institute (CLSI 2019) by comparing 10% DMSO negative control and ciprofloxacin positive control. Antibacterial activity of ethanol extract concentrations of 25%, 50% and 100% obtained an inhibition zone of 8.8 mm, 8.9 mm, and 10 mm. The results of the inhibition zone diameter of the n-hexane fraction with concentrations of 25%, 50%, and 100% obtained an inhibition zone of 6 mm, respectively. The results of the inhibition zone diameter of the ethyl acetate fraction with concentrations of 25%, 50%, and 100% obtained inhibition zones of 18.9 mm, 19 mm and 21.9 mm. The results of the diameter of the inhibition zone for the concentration of water with a concentration of 25%, 50%, and 100% obtained an inhibition zone of 8 mm, 7 mm and 10 mm. The results of the inhibition zone diameter of 10% DMSO negative control concentrations of 25%, 50%, and 100% obtained an inhibition zone of 6 mm, respectively. The results of the inhibition zone diameter of the positive control ciprofloxacin obtained an inhibition zone of 39.1 mm. Pseudomonas aeruginosa is said to be sensitive to antibiotics if it is included in the susceptible criteria, and said to be resistant if it is included in the intermediate and resistant criteria by comparing 10% DMSO negative control and ciprofloxacin positive control. The results showed that ethyl acetate fraction was the most optimum fraction because it could inhibit the growth of Pseudomonas aeruginosa bacteria and had an inhibition zone> 21 mm and was included in the sensitive category.
