Abstract

Black cumin seeds (Nigella sativa L.) have ingredients such as essential oils that can be used as an antidiabetic. Black cumin seed extract is formulated into transdermal patches to avoid first pass effects and maintain drug bioavaibility, in addition essential oils have low solubility so that transdermal patch are made to increase bioavaibility. This study aims to determine the stability of the best transdermal patch preparations for black cumin seed extract and to determine the effect of variations in the concentration of HPMC on the physical stability test of transdermal patch preparations for black cumin seed extract. Black cumin seeds were extracted using the maceration method with 95% ethanol. Black cumin seed extract was made into transdermal patch preparations using HPMC polymer with a concentration of 6%, 7%, and 8%. The bases of patches obtained were subjected to basic testing including pourability and drying time tests, while the obtained patches were tested including organoleptic, weight uniformity of patch thickness, folding resistance, percentage of water content, percentage of elongation, pH and acceptability. Based on research, the black cumin seed extract formulation on transdermal patch preparations showed that the addition of HPMC had the effect of increasing the weight, thickness, pH and elongation percentage of the Transdermal patch with a 6% HPMC concentration that had better physical stability than other formulas with a patch weight of approximately 1,34 g ± 0,03, the patch thickness is 0,03 mm ± 0,00, and the water content is 0% ± 0,00.
