Abstract

Ocimum basilicum leaves is a plant that contains essential oils and phenolic compounds that have the potential to be developed as antibacterial. The most important content in basil is the essential oil found in the leaves and the parts that grow above the ground. This study aims to determine the antibacterial activity of the ethyl acetate fraction of basil leaves against Pseudomonas aeruginosa and Staphylococcus aureus. Ocimum basilicum leaves fraction is the result of fractionation of ethanol extract of basil leaves obtained by suspension using water and then fractionated using liquid-search fractionation (FCC) method. The ethyl acetate fraction obtained was then tested for antibacterial activity with concentrations of 25%, 50% and 100% using the disc diffusion method. In this study, a positive control of 5µl ciprofloxacin and a negative control of 10% DMSO was used. The results showed that the ethyl acetate fraction could inhibit the growth of Pseudomonas aeruginosa and Staphylococcus aureus bacteria, but it was significantly different from the positive control of ciprofloxacin. The ethyl acetate fraction of Ocimum basilicum leaves which gave the greatest inhibition zone against Pseudomonas aeruginosa was at a concentration of 100% with an average inhibition zone diameter of 18.9 mm and the greatest inhibition zone against Staphylococcus aureus was at a concentration of 100% with an average diameter of the inhibition zone of 14.13 mm.
