Abstract

Thyme herb (Thymus vulgaris L.) is a plant that has a lot of phenolic content. The research was carried out to the fraction stage so that the compound distribution would occur. This study aim to determine the phenolic content and characterization of the extract and fraction of thyme herb. Thyme herbs were macerated using 70% ethanol, evaporated using a rotary evaporator, and then fractionated using three solvents, n-hexane, ethyl acetate and water. Identification of phenolic compound was carried out by tube test, TLC, UVVis spectrophotometry and FTIR. Determination of phenolic content was carried out using UV-Vis spectrophotometry using the Folin-Ciocalteu method. The preliminary test of thyme herbs extract and fractions showed positive results with FeCl3 and K2Cr2O7 reagents. The TLC test showed that the extract and the fraction had the same hRf value as the standard, 87.5. UV-Vis spectrophotometric identification showed that thymol, ethanol extract, n-hexane fraction, ethyl acetate fraction and water fraction have lamda max which is 278 nm, 284 nm, 243 nm, 260 nm and 264 nm (polyphenolic compounds 240 - 285 nm). FTIR identification shows that the sample is positive contains fenol because it has the typical functional groups of OH, aliphatic CH, aromatic CH, C=O, C-O-C, aliphatic C=C, and aromatic C=C. The result of the determination of the phenolic content contained in the ethanol extract, n-hexane fraction, ethyl acetate fraction and water fraction were 1.7660 ± 0.0044 %TE; 0.3609 ± 0.0011 %TE; 1.2218 ± 0.0022 %TE; dan 2.2547 ± 0.0022 %TE.
