Abstract
Bacterial growth depends on the source of nutrients. Nutrient agar (NA) is a medium used for bacterial growth. The use of nutrient agar is expensive. The use of alternative media that is cheap, easy to obtain, and has a large source of nutrients for bacterial growth is urgently needed. Black soybeans (glycine soja) have protein and carbohydrate values. This high protein and carbohydrate source is thought to be used in the form of alternative media that supports the growth of Staphylococcus aureus bacteria. This type of research is experimental with Post Test Only Control Design. They subjects of this study were black soybean media (glycine soja) with concentrations of 4%, 6% and 8% which were tested on Staphylococcus aureus bacteria and the total plate number (ALT) was calculated. Based on the results of research conducted, the morphology of Staphylococcus aureus nutrient agar (NA) is the same size as black soybean medium, small in size, golden yellow in color and round in shape. The result of calculating the total plate number (ALT) from nutrient agar (NA) media is 99 X (106) CFU while the total plate number (ALT) from black soybean (glycine soja) media with a concentration of 4% is 47 X (106), soybean media black soybean (glycine soja) with a concentration of 6% was 64 X (106), black soybean medium (glycine soja) with a concentration of 8% was 73 X (106). From the results of this study, it can be concluded that black soybean (glycine soja) can be used as an alternative medium for the growth of Staphylococcus aureus bacteria with the best concentration of 8%.
