Abstract
Background: fixation (preservation) is one of the histotechnical stages that stabilizing important element on the tissue so the element are not dissolved, moved or distorted during further procedures. Time is an important factor in the fixation process, if the fixation less than 1 hour it will cause the tissue to not be colored properly, while fixation more than 24 hours the result is tissue shrinkage. Objective: to determine the effect of the duration of fixation on the microscopic representation of kidney and liver tissues with Hematoxylin-Eosin staining. Method: this study uses literature study method with descriptive data analysis. Results: The results of the study showed that the tissue by fixation within 6-24 hours showed a good microscopic representation of the tissue, while on the tissue by fixation within 1 week or more showed an deficient microscopic representation. Conclusion: based on that result of this study can be seen that the effect of time fixation on the microscopic tissue representation, the longer of fixation time, will be more worse to the microscopic result.
