Abstract
Quercetin is a flavonoid compound in the flavonol group found in the telang flower (Clitoria ternatea L). Quercetin has a potential pharmacological activity, so it needs to be determined. Sample preparation was carried out by maceration extraction technique with 70% ethanol solvent. The HPLC method developed for the determination of quercetin content in the telang flower extract has been optimized in advance with various comparisons of methanol:aquabides as a mobile phase with parameters of resolution, tailing factor 10%, and retention time. The research design is descriptive study. The HPLC systems used are reverse phase HPLC, octadesil column (C18) size 250 mm x 4,6 µm, methanol mobile phase: aquabides optimized at a ratio of 40:60; 50:50; 60:40, flow rate of 1 mL / min, and detector at 371.5 nm wavelength. The optimized HPLC system was used to determine the quercetin content in the telang flower extract. The optimal phase of motion obtained in this study was a mixture of methanol:akuabides (50:50) with a raw resolution result of 2.152 kuersetin and 1.453 sample kuersetin, 10% raw kuersetin 1.127 and 1.031 sample kuersetin, as well as a relative retention time of 0.953. The rate of kuersetin in the late flower extract obtained in this study was 0.078±4.358x10 -3 %. The HPLC method used to determine the levels of quercetin in the telang flower extract in this study was able to produce a coefficient of variation 5.58%, correlation coefficient 0.999, specificity with a resolution between 1.272 to 3.341, limit of detection 0.045 ppm, and limit of quantification 0.150 ppm.
