Abstract

Binahong leaf extract (Anredera cordifolia (Ten.) Steenis.) contains flavonoids, phenols, vitamin C. Binahong leaf extract needs to be made in a dosage form to facilitate its use, one of which is in the form of a gel dosage form. The aims of this study were to determine the ability of binahong leaf extract to reduce free radicals, to determine the optimal ratio of HPMC and Carbopol in the gel formula and to determine the potential antioxidant activity of the gel preparation of binahong leaf extract (Anredera cordifolia (Ten.) Steenis.). The antioxidant activity test of the ethanol extract of binahong leaves was carried out using the ABTS method using a uv-vis spectrophotometer. Optimization of binahong leaf extract gel preparation with a concentration of 0.4% with a combination of gelling agent HPMC and Carbopol determined by Design Expert 11 software using Simplex Lattice Design. The responses used are dispersion, adhesion, viscosity and pH. Verify the optimum formula with One Sample T-Test on SPSS. The optimum gel formula was tested for antioxidants using the ABTS method. The results of phytochemical screening showed that the ethanol extract of binahong leaves contained flavonoids, phenols, vitamin C, alkaloids, and saponins. The results of antioxidant testing on the extract have antioxidant activity with an average IC50 value of 18.85±0.29 which is classified as very strong. The results of the optimization of the binahong leaf extract gel preparation, obtained a comparison of HPMC and Carbopol of 4.22 gram : 0.78 grams with a desirability value of 0.789. The dispersion test value is 4.32±0.014, the adhesion test is 1.38±0.016, the viscosity test is 330.00±8.16 and the pH test is 5. The test results of the weight test value between the optimal formula and the prediction of Simplex Lattice Design with One Sample T-test showed no significant difference, which means it is valid. The results of antioxidant testing on the gel have antioxidant activity values with an average IC50 value of 50.67±0.46 which is quite strong.
