Abstract

Tuberculosis is a direct infectious disease caused by the TB bacteria, named Myobacterium tuberculosis. Until now, Tuberculosis (TBC) is still a big challenge for the Indonesian government. For the purpose of diagnosis by direct microscopic examination of sputum, suspected TB patients are examined for sputum samples with Ziehl Neelsen staining. However, in some areas in Indonesia, the distance from health facilities sometimes requires that Acid-Fast Bacilli samples undergo a delay in examination because they go through the delivery process first. Therefore, the purpose of this study was to determine the effect of differences in temperature and storage time on the amount of Acid-Fast Bacilli +. The design of this study is a laboratory experimental research design with a completely randomized design. The study was conducted with 5 treatments, that stored for 0 hours, 24 hours at a temperature of 2-8oC, 24 hours at a temperature of 25oC, 48 hours at a temperature of 2-8oC and 48 hours at a temperature of 25oC. After microscopic observation and data processing, the results obtained p value > 0.05, then Ho is accepted and Ha is rejected. From these results it can be concluded that there is no effect of differences in temperature and storage time on the amount of acid-fast bacilli +.
