Abstract

Pseudomonas aeruginosa is a bacterium that has a characteristic growth medium that can distinguish it from other bacteria, namely pigments. Factors that influence pigment formation are the source of nutrition for each isolation medium and the incubation time. The purpose of this study was to determine the formation of Pseudomonas aeruginosa pigments when incubated in various isolation media with various time variations. This research uses descriptive design. This research was conducted at the National STIKES Bacteriology Laboratory and the research was conducted in January-May. The samples of this research were Pseudomonas aeruginosa bacteria culture, Tryptic Soy Agar (TSA), Mueller Hinton Agar (MHA), Nutrient Agar (NA), and Brain Heart Infusion (BHI) isolation media in incubation with variations of incubation time of 8, 24, 48, 72 and 75 hours. The sampling technique used is Quota Sampling. The results showed that the Tryptic Soy Agar (TSA) media was the best medium to use compared to the NA, MHA and BHI isolation media for the formation of Pseudomonas aeruginosa pigments with an incubation time of 24 to 72 hours. It can be concluded that each isolation medium used in this study showed differences in the formation of Pseudomonas aeruginosa pigments when incubated with different time variations.
